I want to make sure that when the application is installed, whether on a local machine or on a network, that it is available to ALL users. I am therefore assuming (correctly?) that I need to set Access control. I click on New, click on Grant Everyone Total control, then click on Browse and attempt to select the destination folder itself, but OK does not become enabled. (I also need to ensure total access to all users for the sub-folders under the destination folder as well.)
I do not know whether it will be installed onto a single machine or on a network, so I do not know whether to select File system or Share.
Could somebody explain this to me please.

You want to select File System, unless you are setting the permissions of a shared folder (which is a network share).
Also, this really depends on the application you have at hand. With a "properly authored" application, you may not need to set any privileges whatsoever - it may just work all out of the box assuming the application only writes to the current user's data folders.
But Set Access Control is there to help for every other case

Many thanks for the reply Michael. For this application, the data has to be stored in a sub-folder of the application folder.
If I click on New, select File system, click on Grant Everyone Total control, then click on Browse and attempt to select the destination folder itself rather than a file within the folder, OK does not become enabled. Should I just type in $TARGETDIR$ into the edit box direct rather than trying to use Browse? The Help indicates that I should be able to select files OR folders.

Yes, just type in the folder name. The browse dialog picks files only.
